So, I finally have my very own Sega Megadrive.

Unfortunately I have no clue how to make it work in conjuction with DVD players and other gadgets that were not invented when the Megadrive was at the height of popularity.

Anyone have any pointers?

I have a megadrive. They are awesome. As far as I know you can only plug them into RF (old-school antenna) connections, not AV. So I'm not sure you can plug it into your DVD player - I'm pretty sure you have to plug it straight into your TV antenna plug-hole.
